How Does the LAL Kinetic Chromogenic Assay Work?

The assay involves the following key steps:

  2. Reaction Initiation: The sample is mixed with LAL reagent containing a chromogenic substrate.
  3. Enzymatic Cleavage: Endotoxins activate a cascade of enzymes in the LAL reagent, ultimately cleaving the chromogenic substrate.
  4. Color Development: The cleaved substrate releases a yellow-colored compound (p-nitroaniline).
  5. Kinetic Measurement: The rate of color development is measured spectrophotometrically at 405 nm.
